What is semaglutide?

Semaglutide is a GLP-1 receptor agonist, which means that it works by mimicking the effects of a natural hormone in the body called glucagon-like peptide-1 (GLP-1). It helps with weight loss in obese patients and reduces blood sugar levels in diabetic patients.

How does it work (mechanism of action) for weight loss?

Semaglutide’s mechanism of action for weight loss is thought to be multifactorial, but primarily involves its effects on the appetite control centers in the brain.

Semaglutide is a GLP-1 receptor agonist, which means that it activates GLP-1 receptors in the brain. These receptors are located in areas of the brain that regulate appetite and food intake, including the hypothalamus and brainstem.

What is the evidence behind Semaglutide use and weight loss?

The New England Journal of Medicine published the results of the Semaglutide Treatment Effect in People with Obesity (STEP) clinical trials, which investigated the efficacy and safety of semaglutide for weight loss in adults with obesity.

In the STEP 1 trial, which involved over 1,900 adults with obesity or overweight and at least one weight-related condition, participants who received semaglutide once weekly had significantly greater weight loss compared to those who received placebo. At week 68,
participants in the semaglutide group had an average weight loss of 14.9% compared to 2.4% in the placebo group.

Similarly, in the STEP 2 trial, which involved over 1,200 adults with type 2 diabetes and overweight or obesity, participants who received semaglutide had significantly greater weight loss and better glycemic control compared to those who received placebo. At week 68, participants in the semaglutide group had an average weight loss of 9.6% compared to 3.4% in the placebo group.

The results of the STEP trials demonstrate that semaglutide can be an effective treatment option for weight loss in adults with obesity or overweight and at least one weight-related condition, as well as in adults with type 2 diabetes and overweight or obesity.
